SRX15493783: NextSeq of Ambystoma mavortium skin microbiome

Design: Two mock community samples were added to the library. Cross-contamination oligos and synthetic gene sequences were added to each sample, and samples were normalized. PCR was used to amplify the 16S rRNA V4 region and ITS1 region and add unique dual index combinations to each sample for bacterial and fungal metabarcoding, respectively. Magnetic bead cleanup was performed following PCR, and amplicon product was pooled. Each sample received two PCR replicates.

Amphibian skin microbiota serve as the first line of defense against cutaneous diseases. This project investigates spatiotemporal and ontogenetic variation of bacterial and fungal communities on the skin of the western tiger salamander (Ambystoma mavortium). Additionally, this study identifies differentially abundant microbes between salamander skin and the environment, predicts the antifungal function of skin bacterial communities, and observes whether predicted antifungal function is correlated with amphibian chytrid fungus.
